Formulation Guidance
Matrix erosion rate in sustained-release formulations is tunable through both guar gum percentage and particle size, and the two interact — finer particle size combined with higher percentage compounds the rate of gel-layer formation, which can slow release further than either variable would alone. Confirm both together during early formulation trials rather than optimizing one and then the other.
In wet granulation, guar gum's binder strength is sensitive to granulation liquid volume and drying conditions. It's worth establishing a granulation end-point (via torque or power-consumption monitoring, for example) specific to guar-gum-bound formulations rather than porting over an end-point calibrated for a different binder system, since optimal moisture content at the end-point isn't necessarily the same across binder types.
For oral suspensions, confirm resuspension force against your target patient population's realistic shake behavior — a suspension that resuspends easily in a lab shake test may not perform the same way for a patient population that shakes less vigorously or less consistently.
Because drug-product formulations carry a deeper regulatory documentation requirement than nutraceutical qualification, request elemental impurity data aligned with ICH Q3D risk assessment alongside the standard Certificate of Analysis and DMF Type IV reference — dietary supplement qualification typically doesn't require this same depth of documentation, so it's worth confirming this is available before scale-up.
